Applications of BBa_K1783001

We coupled Hok/Sok to a constitutive RFP reporter found here https://parts.igem.org/Part:BBa_K1783003 . We tested Hok/Sok's ability to maintain the pSB1C3::Hok/Sok::RFP plasmid by growing cells in liquid culture with or without chloramphenicol and determining the proportion of chloramphenicol-resistant cells by plating a dilution of our liquid culture on chloramphenicol plates. At each 24 hour mark, we subcultured into fresh media, and continued plating and subculturing until near complete loss of fluorescence. See results here: http://2015.igem.org/Team:UMaryland/Results . We found that Hok/Sok was able to maintain the plasmid for a long period of time.
Further, our growth curve indicated no retardation in growth when DH5alpha carries a plasmid maintained by Hok/Sok. This is likely because of the low rate of transcription and translation associated with the Hok/Sok machinery.
